Using the classical Parashari method — your Life Stone is the gem of your ascendant (Lagna) lord, your Fortune Stone is the gem of your 9th-house (luck) lord, and your Benefic Stone is the gem of your 5th-house lord. These are the most auspicious planets to strengthen for your chart.

Birth time gives the most accurate ascendant (and Life Stone). If you don't know it, the Fortune and Benefic stones derived from your Moon chart remain reliable, and the Life Stone is given as an approximation.
